In the Three-City prospective cohort study among postmenopausal women over 65 years, plasma estrogens were measured and five common polymorphisms of ESR1 and ESR2 were genotyped. Overall, the risk of arterial ischemic disease significantly increased with estrogens levels after allowance for traditional cardiovascular risk factors . This association was observed in women with ESR1 rs9340799-AA genotype but not in women with AG/GG genotype. The relation was partly mediated through elevated fibrinogen levels and enhanced in vitro thrombin generation . ESR1 rs9340799 genotype may modify arterial risk related to high endogenous estrogens levels in older postmenopausal women and hypercoagulability may act as a mediator.
